Medicago sativa, alfalfa (also known as lucerne) is a perennial herbaceous legume. Lines J101 and J163 contain the epsps gene from Agrobacterium tumefaciens strain CP4 to allow for the use of glyphosate, the active ingredient in the herbicide Roundup®, as a weed control option.

A stacked line of alfalfa with tolerance to glyphosate through introduction of epsps genes from Agrobacterium tumefaciens strain CP4.
Information on the inserted DNA sequences
The T-DNA portion of this plasmid vector contained one cp4 epsps gene expression cassette coding for glyphosate tolerance. The cp4 epsps gene was linked to a chloroplast transit peptide (cpt2) sequence, derived from the Arabidopsis thaliana epsps gene. The expression of the cp4 epsps gene was regulated by an enhanced 35S promoter from the Figwort Mosaic Virus, and the petunia heat shock protein intron (HSP) 70 5’ untranslated leader sequence. The transcriptional termination sequences were derived from the pea (Pisum sativum) ribulose-1,5-biphosphate carboxylase (Rubisco) small subunit E9 gene.
Vector information
Plant transformation was achieved with the plasmid vector PV-MSHT4, a binary vector which contains a region of DNA (T-DNA), which has one cp4 epsps expression cassette under the control of a constitutive promoter flanked by left and right border sequences which is approximately 3.8 Kb. No additional elements from the transformation vector PV-MSHT4, linked or unlinked to the intact gene cassette were detected in the genome of events J101 and J163.

The genetically engineered plant contains a form of the plant enzyme 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) that allows it to survive the otherwise lethal application of glyphosate. The EPSPS gene that was introduced into J101 and J163 was isolated from the common soil bacterium Agrobacterium tumefaciens, strain CP4 and the form of EPSPS enzyme produced by this gene is tolerant to glyphosate.
This plant is a product of traditional plant breeding, and therefore is not automatically subject to regulation in all jurisdictions as are transgenic plants resulting from recombinant DNA technologies. Certain jurisdictions may request notification in advance of the release of a stacked hybrid, or may request information to conduct an environmental and food safety assessment.
